Hepatoprotective and antioxidant effects of Ocimum basilicum extract in CCl4-induced rats hepatotoxicity compared with silymarin.

Abstract

The present study was designed to investigate the hepatoprotective and an-tioxidant effect of Ocimum basilicum extract (OBE) on CCl4 induced hepatotox-ic rats compared with silymarin. Animals were divided into 6 groups. Group 1 was used as a control and received distilled water (1 ml/kg, p.o) daily for 21 days. Group 2 (silymarin administered rats) received silymarin (100 mg/kg, p. o.) daily for 21 days. Group 3 (ocimum administered rats) received OBE (400 mg/kg, p. o.) daily for 21 days. Group 4 considered as (CCL4 injected rats) re-ceived CCl4: corn oil (1:3) 2,5 ml/kg, s. c. on 15th day, 18th and 21st day. Group 5 (CCL4 injected rats protected with silymarin) received silymarin as group 2+CCL4 as group 4. Group 6 (CCL4 injected rats protected with ocimum) re-ceived OBE same to group 3 and CCL4 as group 4. Blood samples were separat-ed into serum for liver functions determination (ALT, AST, GGT, ALP and bili-rubin) and Liver tissue sample for antioxidant parameters determination (CAT, SOD, MDA and GPX) and Immunological assay (TNF, IL-4 and NFkβ). The results revealed that in group 2 there were significant increases in ALT, AST, GGT, ALP, bilirubin, MDA, TNF, IL-4 and NFkβ, while parameters CAT, SOD, and GPX showed significant decrease. Meanwhile, administration of Si-lymarin and Ocimum basilicum extract resulted in significant decrease in all ele-vated mentioned parameters and increase in CAT, SOD and GPX. Therefore, it could be concluded that Ocimum basilicum extract has hepatoprotective and an-tioxidant role but silymarin has significant antioxidant effect than ocimum.
