Evaluation of the Potential Therapeutic Anti-inflammatory, Anti-proliferative and Apoptotic Effects of Viburnum tinus Leaves Different Extracts.

Abstract

Viburnum tinus (V.tinus) is an Adoxaceae family medicinal plant rich in vital phytochemicals such as polyphenols, triterpenoids, flavonoids, coumarins, and iridoids. This study shows that V. tinus leaf extracts have anti-inflammatory, anti-cancer, and apoptotic properties. The plant leaves were dried and soaked in 90% MeOH to prepare the crude extract, which was fractionated by n-Hexane, CH2Cl2, and MeOH. Phytochemicals were detected in these extracts, and their anti-inflammatory, anti-cancer, and apoptotic activities were evaluated in-vitro.The highest content of flavonoids, tannins, terpenoids, and coumarins in the n-Hexane extract stimulates its anti-inflammatory activity, as evidenced by protein denaturation inhibition (94.60±3.90 percent), proteinase inhibition (88.5±0.50 %), membrane stabilization (73.2±1.58 %), and inflammatory cytokine (INF- ɤ) inhibition (100±0.00 %) without any toxicity on blood. It also has a cytotoxic impact on HeG2, MCF7, and CaCO2 cell lines, with IC50 (29.7 2.58, 75.3 4.70, 29.1 0.59 µg/ml) and SI (10.2, 10.4, 4) on HeG2, MCF7, and CaCO2, respectively. The apoptotic impact of n-Hexane in HepG2 resulted in quantitative expression of the BAX gene, which increased significantly and the BCL2 gene, which decreased significantly.
