Abstract

ELISA technique is the most sensitive and specific serological method for diagnosis of fascioliasis. In this study, three tested Fasciola gigantica antigens were used in immunization of three different groups of rabbits to evaluate their diagnostic value using ELISA technique. Those antigens are F.gigantica crude antigen, F.gigantica excretory secretory (E/S) antigen and F.gigantica egg antigen. Antibodies produced by vaccination with E/S antigen proved to be the most specific as they were the least in cross-reaction. While, antibodies produced by vaccination with crude antigen were the most sensitive as they gave the highest ELISA O.D. values. The immune response began from the second week against the target antigen, increased to the maximum values in the fourth week and began to decrease at the sixth week. Using different serum dilutions was a method for increasing the accuracy of serological tests and minimizing the level of cross-reaction between the tested antigens.
