Abstract

The interaction and associated correlation between defect in MMR genes expression, P53 over expression and c-erbB2 over expression on top of HCV associated chronic hepatitis and cirrhosis has a major role in the induction of HCC.This study was conducted on 30 patients with HCC and 30 patients with HCV associated chronic hepatitis. Serum samples & liver tissue samples were taken from all patients.All liver tissue samples were examined to detect expression of MMR genes, (mainly GTBP6, hMLH1 and hMPS I), using multiplex RT-PCR technique and expression of P53 and c-erbB2 genes using immunohistochemical analysis. Our results showed that 56.7% of HCC cases, 6.7% of CAH cases and 20% of ANAPCLT cases and an over expression of P53 gene, while 70% of HCC cases, 56.7% of CAH cases and 80% of ANAPCLT cases had an over expression of c-erbB2 gene and 33.3% of HCC cases, zero percent of CAH cases and 3.3% of ANAPCLT cases had a combined over expression of P53 gene and reduced expression of GTBP6 gene, while 40% of HCC cases, 23.3% of CAH cases and 20% of ANAPCLT cases had a combined over expression of c-erbB2 with reduced expression GTBP6.
