Abstract

Introduction and Aim of the Work: Chemotherapy with cisplatin can have adverse effects on spermatogenesis, so this work aimed at investigating the protective effects of ginger against cisplatin-induced testicular toxicity in male albino ratsMaterials and Methods: Twenty four male albino rats were used in this study. They were divided into three groups; the first served as control, the second was injected by cisplatin (12 mg/kg once) and the third was given ginger (310 mg/kg orally ) for 26 days then injected by cisplatin (12 mg/kg once) at day 21. Testicular specimens were processed for light microscopic examination using H&E and PAS stains. Other specimens were processed for electron microscopic examination.Results: In cisplatin administered group; by light microscope; there were distortion and shrinkage of seminiferous tubules, depletion, vacuolation and degeneration of spermatogenic cells with exception of spermatogonia which resist cisplatin toxicity. In addition to thickening and separation of basement membrane from overlying cells. Ultrastructurally; there were distortion, shrinkage of some spermatogenic cells, degeneration of cytoplasm. Also, small condensed nuclei with ill defined nuclear membrane were observed. In ginger and cisplatin group, there was partial protection as revealed by light and electron microscopic studies inspite of some degenerative changes. Statistical analysis of the obtained results revealed presence of a significant decrease (P ≤ 0.001) in body and testicular weights, diameter of seminiferous tubules and number of normal tubules in cisplatin group when compared to control group. Meanwhile , there was a significant increase (P ≤ 0.001) in body and testicular weights, diameter of seminiferous tubules and number of normal tubules in ginger and cisplatin group when compared to cisplatin group.Recommendations: The use of ginger has some protective effects; so more experiments with increasing the dose or prolongation of administration period could be beneficial for patients taking chemotherapeutic drugs.
