Objecitve : To assess the effect of hypothermia (20C and 28C) and vascular perfusion with two types of preservation solution (UW and HTK) to decrease the I/R injury of the liver. Methods : Pig livers are perfused in situ with a chilled solution (4˚C) attaining tissue temperatures of (either 28˚C or 20˚C) during 120 minutes of (TVE). Ringer-Glucose, HTK and UW are used for perfusion. There was no perfusion in control. Results : Survival was 6/6 in all groups except in control group was 4/6. Plasma AST, Exogenous HA uptake, Tissue glutathione and Histopathology score were significantly better in perfused groups versus control group and better in UW perfused groups versus the other perfused groups. Bile production, ICG, Intrahepaic tissue pO2, IL6, TBARS were significantly better in perfused groups versus control group, but there was no significant differences between different perfusion solutions. No significant differences between 28˚C and 20˚C perfused groups were found. Conclusion : Cooling of the liver to 28˚C or 20˚C by in situ hypothermic perfusion during 120 minutes of ischemia significantly protects the liver from I/R injury. UW perfused groups showed significantly better results in relation to other perfused groups.
