One hundred and fifteen men were included, divided into : control (n=14), OAT with varicocele (n=17), OAT (n=18), AT with varicocele (n=10), AT (n=10), OA (n=10), 36 NOA cases [TESE+ve(n=26),TESE-ve (n=10)]. Azoospermic cases were subjected to testicular biopsy for histopathology and diagnostic TESE. Seminal plasma (SP) samples were subjected to polyacrylamide gel electrophoresis (SDS-PAGE) and estimation of amino acid by paper chromatography. Results showed that individual SP proteins contribution is similar for each individual, whatever the andrological condition. SDS-PAGE of SP proteins, as well as non-essential amino acids could not offer a reliable method for predicting successful sperm retrieval. SP aminogram of essential amino acids; histidine & lysine followed by valine, total content, leucine & isoleucine, can offer a simple method for predicting successful sperm retrieval. Also, SP aminogram of non-essential amino acids; total content, arginine and tyrosine can offer a reliable method to differentiate OA and NOA.
