The purpose of this study was to detect the effect of khat extract on the rat submandibular and sublingual salivary glands through histological and electron microscopic examination as well as immunohistochemical localization of Fas protein. The study was carried out using thirty healthy adult male albino rats of average weight 150 ± 10 grams. The rats were divided into two main groups; control group which included untreated animals and experimental groups which subdivided into two groups.Experimental group (І): received 1 ml of khat extract twice a week for 5 weeks via oropharyngeal tube and experimental group (ІІ): received the same dose for 10 weeks.The glands of one side were prepared for light microscope examination using H&E and Masson trichrome stains and the glands of other side were prepared for electron microscopic examination. Also, some specimens stained immunohistochemically for Fas protein.Conclusions: Administration of khat extract produced moderate degenerative changes in the histological and ultrastructural of rat submandibular and sublingual salivary glands. Paired Student t-test revealed that there was a highly significant difference in mean Fas optical density between control and experimental group. And the ANOVA analysis showed a highly significance difference between different groups.
