Abstract

Background: Acrylamide became an important public health concerns as it is detected in widely consumed food items as fried bread, potato chips, and any carbohydrate-rich food cooked at high temperatures. Because of many health hazards induced by its chronic consumption. Various health organizations recommend monitoring its presence in certain food. Aim of the work: To investigate the effect of the abrupt and gradual withdrawal of acrylamide on its induced hepatic and renal cortical injury of adult male albino rats. Materials and methods: Rats were divided into 4 groups 10 per each group. The control group (group I), received water and standard diet orally. The acrylamide group (group II): received acrylamide at the dose of 20 mg/kg orally via intra-gastric gavage route once daily for 6 weeks. The abrupt withdrawal group (group III): received acrylamide at the dose of 20 mg/kg once daily for 6 weeks then abruptly stopped administrated in the following 3 weeks. The gradual withdrawal group (group IV): received acrylamide at the dose of 20 mg/kg once daily for the first 6 weeks. In the following three weeks, acrylamide was gradually withdrawn. Livers and renal cortical tissues were removed. They were used for paraffin embedding for histological (H&E, PAS and Masson trichrome), immunohistochemical (anti- PCNA, caspase 3 and alpha smooth muscle actin antibodies) and morphometrically studies. Results: The histological, immunohistochemical and morphometric results showed tissue sections of liver and renal cortex of groups II and IV appeared with marked destruction of the histological architecture in the form of cellular apoptosis, vascular congestion and inflammatory cellular infiltrations. While the group III tissue sections revealed restoration of their histological architecture. Conclusion: The administration of acrylamide caused many morphological changes in both liver and renal cortical tissues due to reactive oxygen species production. The sudden withdrawal of acrylamide made marked improvement if compare with the gradual withdrawal which led to minimal or no improvement. Recommendation: We must adopt a healthy eating plan by replacing cooking for longer periods or at higher temperatures by boiling and steaming in cooking food. Consumers should also not hesitate to stop using acrylamide containing products suddenly. Further researches will be needed to looking for natural healthy foods contain the antioxidants to protect against the damage induced by these daily used products.
