Alleviation of Tissue Browning During Clonal Propagation of Banana cv. Grand Naine

Abstract

Banana micropropagation is commercially established by the use of shoot tips obtained from intact plant on shoot multiplication medium under optimal culture conditions. However, this is somewhat associated with tissue browning, which may be associated with a reduced efficiency of shoot multiplication process. An increase or decrease in incubation temperature from 27 ºC, change the texture of the media from semisolid state, addition of ascorbic acid and substitution of optimal concentration of benzyl amino purine (5 mg/l BAP) with 2-isopentyl adenine (4 mg/l 2ip), decreased shoot multiplication and growth, even if they reduced tissue browning. Our results indicated that the chemical composition of the MS medium containing 5 mg/l BAP was ideal for banana shoot multiplication. Addition of some natural ingredients rich in many important components (yeast extract, coconut endosperm milk or moringa leaf extract) was unsuitable for in vitro multiplication of banana. The observed exception was that growth parameters of shoots formed on MS medium with 4 mg/l 2ip were better than those formed on MS with 5 mg/l BAP. In addition, shoot multiplication on 2-isopentenyl adenine (2ip) containing media showed root formation, which facilitated the adaptation and transfer of plants to soil.
