The Potential Therapeutic Effect of Cerebrolysin against Cyclosporine A - Induced Cerebellar Toxicity in Adult Male Albino Rats

Abstract

Background: Cyclosporin A is an immunosuppressive agent used to treat organ rejection post-transplant and autoimmune diseases. It can induce toxic effects on the cerebellar cortex such as cerebellar ataxia, vasogenic edema and occasional supratentorial lesions. Cerebrolysin possess therapeutic strategies as a neurotrophic factor with neuroprotective effects. Aim of the work: Evaluation of potential therapeutic cerebrolysin's effect on cerebellar toxicity induced by cyclosporine A in adult male albino rats. Methods:40 adult male albino rats were randomized into 4 groups and received all treatments for 4 weeks. Group I, served as control. Group II, cerebrolysin was given a daily intraperitoneal injection of 2.5 ml/ kg. Group III received cyclosporin daily with a dosage of 25 mg/kg/day dissolved in olive oil, orally by gavage. Group IV, Cyclosporin A concomitant with cerebrolysin were administered. After 4 weeks, animals were sacrificed by decapitation. Hematoxylin and Eosin staining was performed on the prepared sections, Cresyl fast violet, Glial Fibrillary Acidic Protein immunostaining, electron microscopic examination and assessment of gait. Results: Purkinje and granule cells appeared irregular with irregular nuclei. Nerve fibers have faint myelin sheath and/ or focal areas of myelin. Less abundant Glial Fibrillary Acidic Protein positive cells and decreased Nissl's granules in perikarya and gait impairment. Cerebrolysin restored most of the histopathological changes and locomotor behavior. Conclusion: Cerebrolysin has an ameliorating effect on cyclosporin A- induced cerebellar toxicity in adult male albino rats.
