PREPRATION OF AN INACT IVATED LUMPY SKIN DISEASE VIRUS VACCINE

Abstract

Lumpy skin disease (LSD) local strain (Ismalia 88) was adapted in MA104 cell line and propagat-ed for seven serial passages to obtain a titer or 10-7 /ml TCIDSO. To determine the optimum ime for harvestation of the adapted virus, sequen-ual follow up of the growth behavior of LSD on MA 104 was carried out by studying the growth curve of the virus in the cells. At 2,12,16,24,48,72,96 and 120 hours post inocula-lon. The virus was inactivated using different concentration of binary ethyleneimine (BEI) in-“luding 0.5, 1.2 and 5%. The inactivated virus Was used to prepare the suitable inactivated LSD local vaccine, Virus preparations with titers of 10°, 107, and 108 TCIDs0/ml were adjuvated with 2 types of adjuvants (alhydragel at a final concen-tration of 30% and maracol oil). The vaccine dif-ferent preparations were characterized using dif-ferent techniques including the adsorption affinity assay, drop test, emulsion viscosity and stability as well as keeping quality. In addition, the vac-cine preparations were tested for the stenlity and safety. Thirty five calves divided into 7 groups were inoculated with the different vaccine prepar-ations to evaluate the vaccine potency. Sera were collected weekly post inoculation for 4 weeks and at 2nd and 3rd and 4th months post inoculation. The sera were tested by virus neutralization test and ELISA to evaluate the immune response against the inoculated LSD vaccine preparations. The results of the in vivo experiment revealed that the double oil emulsion vaccine preparation elicited. the highest antibody response aganist LSD at 8 weeks post vaccination whereas the alhydragel vaccine preparations yielded the highest antibodies response at 4 weeks post vaccination.
