PREPARATION OF DNA VACCINE AGAINST INFECTIOUS BURSAL DISEASE VIRUS AND EVALUATION OF ITS IMMUNIZING

Abstract

The efficacy of genetic immunization was investigated by direct DNA inoculation into chickens of gene constructs coding for the protective anti- gens of the Infectious bursal disease virus (IBDV). The VP2, VP4 and VP3 genes of IBDV virulent strain 52/70 were cloned in an expression vector. Chickens were vaccinated by inoculation of 800mg (400mg/dose at two weeks interval) of DNA using a combination of three routes; intramuscular (i.m.), intraperitoneal (i.p) and intradermal (i.d.). Another positive control groups included chickens vaccination of with the intermediate live attenuated D-78 (intraocular) and inactivated 52/70 oil emulsion (intramuscular) vaccines of IBDV. Negative control groups included a group of chicken that was injected with the parent control plasmid and another group that remained untreated. An additional group of mice was inoculated with the same dose of the prepared DNA vaccine and parent control plained Sera obtained from all vaccinated and the non-vaccinated chickens were examined for determination of anti-IBDV specific antibodies using enzyme linked immunosorbent assay (ELISA) and agar gel precipitation (AGPT). While, those obtained from mice were examined by AGPT only. The obtained results revealed that DNA immunization stimulated synthesis of IBDV-specific antibodies. Also, challenge experiment using virulent IBDV strain 52/70 indicated the capability of the prepared DNA vaccine to stimulate high protective immunity in the immunized chicken.
