Abstract
In the present study, a total of fifteen urine samples from bladder cancer patients, were used for isolation of bacterial isolates and these isolates were subcultured on a specific medium for production of pycocyanin pigment. One isolate was selected and identified using morphological, physiological and biochemical methods. The results were confirmed using API 20 specific media. It was found to be Pseudomonas aeruginosa M20. Different media (Mineral salt - methanol medium and Trypticase soy broth medium) were used for Vitamin B12 production. In addition, different solvents (Methanol –water and acetone) were used for extraction of vitamin B 12. Trypticase soy broth and acetone were the most probable medium and solvent for Vitamin B12 production. Two doses of gamma irradiation were used (1 and 2 kGy) to study its effect on vitamin B12 production which showed positive result and effect of gamma irradiation on the ant microbial activity of the extract against the bacterial and fungal strains. Moreover, HPLC, IR and EDX were used for the characterization of the bacterial extract. The results were compared with commercial vitamin B12.The antimicrobial activities of both irradiated and non-irradiated bacterial extracts against bacterial and fungal strains were tested.
