Abstract

Date palm somatic embryogenesis has become an ideal technique for large scale production of distinguish and rare cultivars. Date palm cultivar, culture medium components and culture conditions have been identified as detrimental factors affecting somatic embryo formation process.  This research investigated the potential impact of ammonium nitrate (NH4NO3), calcium nitrate Ca(NO3)2 strengths and polyethylene glycol (PEG) concentrations on mature, germinated and vitrified embryos during maturation stage of cv. Medjool callus cultures. Also, the effect of calcium chloride (CaCl2.2H2O) levels that could improve somatic embryo germination (shoot formation) and secondary embryo numbers during germination stage was examined. Rooting was also determined as affected by phloroglucinol (PG) levels. The ideal amount of potassium sulphate (K2SO4) or aluminosilicate (Al4Si4O10 (OH)8) (kaolin) as anti-transpirants that influenced the survival and growth of cv. Medjool plants was also investigated in relation to the ex vitro acclimatization conditions. Modification of Woody Plant Medium (WPM) maturation medium by using NH4NO3 at double strength and Ca(NO3)2 at fourth strength encouraged the highest significant value of mature embryos formation. NH4NO3, Ca(NO3)2 and PEG had a clear impact on mature, germinated and vitrified embryos during maturation stage of cv. Medjool callus cultures. Also, CaCl2.2H2O levels improved somatic embryo germination (shoot formation) and secondary embryo numbers during germination stage. PG in the rooting medium had a beneficial role in root system. Using Al4Si4O10 (OH)8 at 0.1 mg/l as an anti-transpirant material enhanced survival percentage of plant growth during ex vitro acclimatization stage.
