Backgroumd:Salivary gland tumors represent (SGT) a significant division of maxillofacial pathology. Pleomorphic adenoma (PA) is the most common benign SGT, which revealed a variation in pathological profile and can show malignant transformation after long time to be known as carcinoma ex pleomorphic adenoma (CEPA).Fascin is a protein responsible for actin bundling which enhances the motility of tumoral cells. D2-40 is an antibody made for recognition of podoplanin which can be found in endothelial cells of lymphatics and has a role in tumor intiation.
Aim of the study:This study was conducted to scope and find out the possible role of both fascin and D2-40 in the biologic behavior of benign and malignant pleomorphic adenomas.
Materials and Methods: A total of 20 formalin paraffin tissue blocks of PA and CEPA were included in this study. Hematoxyline and eosine sections were done to confirm diagnosis. Both of lesions were immunohistochemically examined for presence of fascin and podoplanin using BenchMarkVentanaautostainer and evaluated by 2 separate pathologists using light microscope. Statistical analysis was performed using using chi square test (X2-value), A P value <0.05 was considered statistically significant.
Results:There was no significant difference of clinical parameters with fascin and D2-40 expression among PA and also CEPA.
There was no significant difference between PA and CEPA according fascin and D-40 expression P value in both antibodies > 0.05.
Conclusion: Fascin and D2-40 are difficult to differentiate between PA and CEPA. It is hard to predict pathological and clinical behavior in PA and CEPA using fascin and D2-40 expression.
