Selective Cytotoxic and Chemoprotective effect of Epigallocatechine-3-Gallate against human HCT-116 cell line

Abstract

Background: Epigallocatechin-3-gallate use as antioxidant and anticancer may related to its high   bioavailability.   Aim: this study was designed to investigate the possible chemoprotective and cytotoxic effect of Epigallocatechin-3-gallate against Human colorectal carcinoma (HCT-116) cell line. Material and methods: MTT assay was performed to detect the cytotoxic effect of Epigallocatechine-3-Gallate against normal and HCT-116 cells, and determination of gene expression of VEGF,BFGF and Caspase-3 was performed to detect the apoptotic effect of Epigallocatechin-3-gallate  Results: Epigallocatechin-3-gallate has a selective cytotoxic effect on human HCT-116 cells with no cytotoxic effect on healthy cells. At the same time, Epigallocatechine-3-Gallate increases the expression of Caspase-3 with reduction in expression of BFGF, VEGF gene .   Conclusion: Epigallocatechine-3-Gallate has selective cytotoxic and chemoprotective effect against human HCT-116 cells according to its dose used.
