ISOLATION AND IDENTIFICATION OF LIPOLYTIC BACTERIA AND PRODUCTION OF LIPASE

Abstract

Microbial lipases have highest potentiality due to their reacton selectivity
and mild conditions. Accordingly, Ihis worX was suggested 10 isolate and identity
some microorganisms capable of lipase production and to optimize the culture
conditions for these microorganisms to produce lipase. The organisms were Isolated
from clayey 5011, and identified as Pseudomona~ aeruginosa, P. fluorescens and
Bacillus cereus. tncubauon periods for lipase production from, P. fluorescens
P. aeruginosa and B. cereus were one day, 2 days and 3 days, respectively. The maximal yield of lipase production from P. ae(!1ginosa, P. fluorescens
and B. cereus was observed at 30°C. The optimal pH value was 6.0. The maximum
enzyme yield was obtained when the production medium was supplied with 1 %
glucose as a carbon source in the presence of olive oil. The highest growth and
lipase production were observed With ol/ve oil as inducer ali % concentration. Three
organic and inorganic nitrogen sources were added separalely to the fermentation
media, peptone plus urea gave maximum production of lipase (10.9 urno: FFA I mi I
min) at nitrogen eN) concentrallon of 0.92% ror P. 8e(!1ginosa, while P. fluorescFJns
recorded 8.50 )J.mol FFN mIl min and B. C&r&us 4.98 urnol FFA J mil min.
