DETECTION OF Brucella melitensis FROM FILTERED BOVINE PERIPHERAL-BLOOD BY POLYMERASE CHAIN REACTION TEST

Abstract

Brucella melitensis contaminants of bovine peripheral-blood can be concentrated and isolated by filtration; and their DNA can be released for subsequent PCR detection by heating the isolated colony at 95°C for 10 min. Detecting rate of brucellosis in the infected sample was less than 20 CFU/ml of blood, after leukocytes filtration and culturing, as well as on isolated colonies. This PCR method also enables us to make the diagnosing of brucellosis for the animals with a negative complement fixation test.
