Abstract

تمَّ عزل ومضاعفة مورثة الزنک بروتياز بواسطة الـPCR  باستخدام الـDNA  المُستخلص من جينوم البروسيلا الضأنية . صُمم زوج من المرئسات لهذه المورثة يحويان  مواقع التقييد لأنزيمي NdeI و BamHI في النهايتين 5' و3' على التوالي. حيث استخدمت تلک المرئسات في مضاعفة شدفة بطول 927 pb. هُضِـمت الشدفة وأُدخلت ضمن بلازميد التعبـير pET-15(b) المقطوع بنفس الأنزيمات، أدخـل البلازميد المؤشب فيما بعد ضمن بکتريا الـ Escherichia coli BL21 (DE3) للتعبـير عن بروتين الزنک بروتياز. تبين بعـد ساعتين من الحث بتراکيز مختلفة من IPTG ، أن التعبير عن هذا البروتياز حرّض الموت الخلوي البکتيري.
