Abstract

Four media with different additives were prepared and used for propagation of Brucella melitensis rev 1 strain for vaccine production and compared with the currently used serum trypticase soya agar medium. All the used nutrient additives gave good results with different degrees, It was found that medium with yeast extract and protose -ME was the best in growing of the strain as measured by packed cell volume .Vaccines prepared from the strain grown on those media showed no different results in potency of Guinea pigs. So this study recommends using yeast extract and protose -ME as additives to give highest packed cell volume without changing strain character.
