CURATIVE EFFECTS OF GARDEN CRESS OR CARDOON WITH SILYMARIN IN TREATMENT OF HEPATIC, RENAL AND TESTICULAR DEFECTS IN RATS INDUCED BY ACETAMINOPHEN

Abstract

Acetaminophen [N-acetyl-p-aminophenol (APAP)] is widely used as analgesic and anti-pyretic drug, however, it's over dosage is known to be hepato-nephrotoxic in human and experimental animals. The current study was designed to assess the hepato-nephrotoxic and testicular dysfunction effects of a single acute dose of APAP (250 mg/kg b.wt.) in rats and speculate which of silymarin (Sily) (100 mg/kg b.wt.), Garden Cress (G.C) (100 mg/kg b.wt.), Cardoon (Card) (100 mg/kg b.wt.) extracts or their co-administration have best mitigating effects against changes in the antioxidant parameters, lipid peroxidation markers and functions of liver, kidney and testis induced by this acute dose. Seventy adult male rats were divided into 10 groups: the 1st (control group); 2nd (APAP group); 3rd (Sily group); 4th (G.C group); 5th (Card group); 6th (APAP+Sily group); 7th (APAP+G.C group); 8th (APAP+Card group); 9th (APAP+Sily+G.C group) and 10th (APAP+Sily+Card group). Rats of APAP group showed high significant increase in ALT, AST, ALP, urea, uric acid, creatinine, malondialdehyde (MDA) and nitric oxide (NO). On the other hand, this group showed high significant decrease in glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), sperm count, normal morphology and motility of sperm and testosterone activity. The changes in these biochemical parameters are associated with obvious histological defects that appear in liver, kidney and testicular tissues. The animals of Sily, G.C or Card groups didn't show any significant changes in all the previous parameters or histological examination. Whereas, the APAP animals-treated with Sily, G.C or Card showed partial ameliorative effect against APAP-induced hepato-nephrotoxicity and testicular effect with convergent degrees except that Sily treatment is better than G.C and Card, respectively. Finally, rats of the 9th and 10th groups showed significant curative effects, however the curative effect of Sily-G.C was remarkably noticed than Sily-Card.
