Cancer therapy using magnetic nanoparticles combined with Methotrexate drug and microwave irradiation

Abstract

Magnetic hyperthermia (MH) treatment, which involves heating tumor tissues to a moderate temperature of 40–45°C, has emerged as an effective strategy for treating tumors. This method is highly efficient at destroying tumor cells and does not induce the side effects of conventional cancer treatments. MH is based on localized heating of tumor with keeping the healthy cells out of harm. In the process, MH involves dispersing hematite and magnetite nanoparticles(Fe2O3&Fe3O4-NPs) throughout the target tissue and then exposed to microwave MW energy to cause the particles to be heated up to the required temperature of about 45°C. Hematite and magnetite nanoparticles were prepared through the co-precipitation method and characterized using TEM, SEM and X-ray diffractometer. Magnetic behavior of nanoparticles was studied using vibrating-sample magnetometer (VSM). Forty-nine male mice injected with 0.2µl of a suspension of 106 cells/ ml isolated from Ehrlich ascites carcinoma (EAC) in mice, mice were randomly divided into the following groups: EAC, Fe3O4-NPs, Fe3O4-NPs, Methotrexate drug (MTX), Fe3O4-NPs@ Fe2O3-NPs @ MTX,MW, Fe3O4-NPs@ Fe2O3-NPs @ MTX with MW hyperthermia.Through the period of treatment, tumor volume was determined, and tumor apoptosis was studied using the histopathology of the treated tissues. The results indicate that the treated of tumor by the magnetic fluid is a suitable mediator for cancer treatment using magnetic nanoparticles combined with MTX and microwave irradiation which revealed cell infiltrations,extensive necrosis, and apoptosis. It was concluded that microwave therapy with MXT drug in the presence of HNPs and MNPs has a possible and clear antitumor effect.
