Cytotoxicity, solubility and wettability of three root canal sealers : Comparative study

Abstract

This study was performed to investigate: cytotoxicity, solubility and wettability of a premixed calcium silicate–based sealer (Smart Paste Bio and MTA Fill Apex), and an epoxy resin sealer (AH Plus Jet). Cytotoxicity was evaluated using MTT assay using BHK cell-line for three different periods (24hr,72hr and 7 days) with different concentrations of sealer (6.25,12.5,25,50 and 100%). Solubility of the selected sealers was evaluated by measuring the intial weight and final weight after immersion in water for a period of 7 days, using 30 teflon moulds measuring 1.5mm in thickness and 7.75mm in width which were divided into 3 groups (n=10). Wettability was measured using contact angle measurement. The droplet meniscus base width and height was shaded using the image analysis software capture (Scope capture) to measure the equilibrium contact angle mathematically The cytotoxicity test results of the present study showed that; as the concentration of sealer increase, the cytotoxicity increased. In the 24 and 72hr period, AH Plus Jet proved to be least toxic followed by Smart Paste Bio and the most toxic was MTA Fill Apex. In the 7th day period, AH Plus Jet and MTA Fill Apex were toxic at 100% concentration and gradually the toxicity decreased till it became slight toxic. The most toxic was the Smart Paste Bio which remained toxic with different concentrations. The solubility test result of this study showed that the least soluble sealer was the AH Plus Jet which was within ANSI/ADA (2000) standards (<3%), followed by MTA Fill apex and Smart Paste Bio. Both of the calcium silicate based sealers were not within the ANSI/ADA since their solubility was more than 3%. The wettability test result showed that all the three selected sealers proved to have good wetting since the equilibrium contact angle was less than 90 °. MTA Fill Apex proved to have the best wetting behavior followed by AH Plus Jet and Smart Paste Bio.
